Scientists discover a brain cell malfunction in schizophrenia

Scientists at The Scripps Research Institute have discovered that DNA stays too tightly wound in certain brain cells of schizophrenic subjects.

The findings suggest that drugs already in development for other diseases might eventually offer hope as a treatment for schizophrenia and related conditions in the elderly.

The research, now available online in the new Nature journal, Translational Psychiatry, shows the deficit is especially pronounced in younger people, meaning treatment might be most effective early on at minimizing or even reversing symptoms of schizophrenia, a potentially devastating mental disorder associated with hallucinations, delusions, and emotional difficulties, among other problems.

"We're excited by the findings," said Scripps Research Associate Professor Elizabeth Thomas, a neuroscientist who led the study, "and there's a tie to other drug development work, which could mean a faster track to clinical trials to exploit what we've found."

A Promising New Field

Over the past few years, researchers have increasingly recognized that cellular-level changes not tied to genetic defects play important roles in causing disease. There is a range of such so-called epigenetic effects that change the way DNA functions without changing a person's DNA code.

One critical area of epigenetic research is tied to histones. These are the structural proteins that DNA has to wrap around. "There's so much DNA in each cell of your body that it could never fit in your cells unless it was tightly and efficiently packed," said Thomas. Histone "tails" regularly undergo chemical modifications to either relax the DNA or repack it. When histones are acetylated, portions of DNA are exposed so that the genes can be used. The histone-DNA complexes, known as chromatin, are constantly relaxing and condensing to expose different genes, so there is no single right or wrong configuration. But the balance can shift in ways that can cause or exacerbate disease.

DNA is the guide that cellular machinery uses to construct the countless proteins essential to life. If portions of that guide remain closed when they shouldn't because histones are not acetylated properly, then genes can be effectively turned off when they shouldn't be with any number of detrimental effects. Numerous research groups have found that altered acetylation may be a key factor in other conditions, from neurodegenerative disorders such as Huntington's disease and Parkinson's disease to drug addiction.

A Good Idea

Thomas had been studying the roles of histone acetylation in Huntington's disease and began to wonder whether similar mechanisms of gene regulation might also be important in schizophrenia. In both diseases, past research in the Thomas lab had shown that certain genes in sufferers were much less active than in healthy people. "It occurred to me that we see the same gene alterations, so I thought, 'Hey, let's just try it,'" she said.

Working with lead author Bin Tang, a postdoctoral fellow in her lab, and Brian Dean, an Australian colleague at the University of Melbourne, Thomas obtained post-mortem brain samples from schizophrenic and healthy brains held at medical "Brain Banks" in the United States and Australia. The brains come from either patients who themselves agreed to donate some or all of their bodies for scientific research after death, or from patients whose families agreed to such donations.

A great deal of epigenetic research has focused on chemical alterations to DNA itself. Histone alterations have been much more difficult to study because such research requires that the histones and DNA remain chemically intact. Many researchers feared that these bonds were disrupted in the brain after death. However, Thomas's group was able to develop a technique for maintaining the histone-DNA interactions. "While many people thought this was lost, we were able to show that indeed these interactions are preserved in post-mortem brain, allowing us to carry out these studies," said Thomas.

Compared to healthy brains, the brain samples from subjects with schizophrenia showed lower levels of acetylation in certain histone portions that would block gene expression. Another critical finding was that in younger subjects with schizophrenia, the problem was much more pronounced.

Need for New Treatment Options

Just what causes the acetylation defects among schizophrenic subjects?what keeps certain pages of the DNA guide closed?isn't clear, but from a medical perspective it doesn't matter. If researchers can reliably show that acetylation is a cause of the problem, they can look for ways to open the closed guide pages and hopefully cure or improve the condition in patients.

Thomas sees great potential. Based on the more pronounced results in younger brains, she believes that treatment with histone deacetylase inhibitors might well prove helpful in reversing or preventing the progression of the condition, especially in younger patients. Current drugs for schizophrenia tend to treat only certain symptoms, such as hallucinations and delusions, and the drugs have major side effects including movement problems, weight gain, and diabetes. If deacetylase inhibitors effectively treat a root cause of the disease and prove sufficiently non-toxic, they might improve additional symptoms and provide a major expansion of treatment options.

Interestingly, some of the cognitive deficits that plague elderly people look quite similar biologically to schizophrenia, and the two conditions share at least some brain abnormalities. So deacetylase inhibitors might also work as a treatment for age-related problems, and might even prove an effective preventive measure for people at high risk of cognitive decline based on family history or other indicators.

Cristina Fernandez, Argentina President, Diagnosed With Thyroid ...

BUENOS AIRES, Argentina ? President Cristina Fernandez has thyroid cancer, but test results showed Tuesday that it remains limited to a lobe in the right side of her neck and has not spread into her lymph nodes, her spokesman said.

Fernandez, 58, will undergo surgery on Jan. 4 and then take 20 days of medical leave, during which Vice President Amado Boudou will run the country. Meanwhile, she will keep up her normal routine, spokesman Alfredo Scoccimarro suggested, noting that she will appear at several events on Wednesday as planned.

Scoccimarro said the cancer was discovered during a routine exam on Dec. 22, and that Fernandez received the results from follow-up tests hours before the announcement.

This kind of thyroid cancer is highly survivable, with more than 95 percent of patients living at least 10 years after detection, according to the U.S. National Institutes of Health.

The usual treatment is to surgically remove as much of the cancerous material as possible, and then follow up with radioactive iodide treatments, taken orally. This substance helps to destroy any remnants of the cancerous gland and provide for clearer images showing any additional cancer, the NIH said on its Web site.

After surgery, patients usually must take medicine ? levothyroxine sodium ? for the rest of their lives to replace a hormone that the thyroid glands produce. Blood tests every six to 12 months to measure thyroid levels also are recommended.

Fernandez is only the latest South American leader to be diagnosed with cancer. Presidents Fernando Lugo of Paraguay, Hugo Chavez of Venezuela and Dilma Rousseff of Brazil all have undergone treatments recently.

Presidential doctors Luis Buonomo and Marcelo Ballesteros said the operation will be led by Dr. Pedro Saco, chief of the surgery department at Hospital Austral and chief of the Head and Neck Service of the oncology institute at the University of Buenos Aires. Saco also trained in cancer centers in Houston and New York, the hospital said.

Thyroid surgery is not without risk: the NIH says a nerve that controls the vocal cords can be damaged, and doctors sometimes accidentally remove the parathyroid gland, which helps regulate blood calcium levels.

Japan looking to buy Chinese government bonds (Reuters)

BEIJING (Reuters) ? Japan will seek to buy Chinese government bonds, the Japanese government said in a statement issued after Prime Minister Yoshihiko Noda held talks in Beijing with Chinese Premier Wen Jiabao on Sunday.

The statement given to reporters gave no details of when Japan might seek to buy the bonds or on what scale.

Japan had earlier indicated it is in talks to buy Chinese government debt to strengthen economic ties.

Despite sometimes rancorous political ties between the two neighbors, Japan's economic fortunes are increasingly tied to China's economic growth and consumer demand.

China has been Japan's biggest trading partner since 2009.

In 2010, trade between the two nations grew by 22.3 percent compared to levels in 2009, reaching 26.5 trillion yen ($339.3 billion), according to the Japan External Trade Organization.

China and Japan are also the world's first and second-biggest holders of foreign reserves.

Almost every channel has a New Year's Eve special to ring in 2012.

The classic, of course, is the clumsily named?"Dick Clark's New Year's Rockin' Eve With Ryan Seacrest." Like Ruth's Chris Steak House, the special's convoluted title?pays tribute to its original host and the new one. (Dec. 31, 10 p.m., ABC)

NBC's New Year's celebration is "New Year's Eve With Carson Daly," which also?features news anchor Brian Williams.

Prefer a down-on-the-farm special? "American Country New Year's Eve" features "American Idol's" Lauren Alaina and other country performers. (Dec. 31, 11 p.m., Fox.)

Movies
Moviegoers are spoiled for choice this holiday.

If it's action you're looking for, "Mission Impossible: Ghost Protocol" turns Tom Cruise into James Bond, with stunning stunts, beautiful women, and Cruise hanging off the world's tallest building. (In theaters now.)

If the whole family wants to see something dramatic but inspirational, "War Horse" is a good bet. Spoiler: Animal lovers and those with sensitive stomachs should know the horse manages to come through World War I pretty much intact, and there's a happy ending. (Opens Dec. 25.)

And fans of Stieg Larsson's Millennium trilogy have long awaited the English remake of "Girl With the Dragon Tattoo." Early fears were that it couldn't live up to the 2009 Swedish-language version, but reviews have been quite positive. (In theaters now.)

DVD
George Clooney plays a grim assassin who's rethinking his career in "The American," a very serious thriller that may give you a completely different view of the handsome actor. (On DVD Dec. 28.)

Their lives still fascinate: Sid Vicious killed Nancy Spungen in 1978, and the Sex Pistol bassist died himself in 1979. "Sid & Nancy" has been a cult favorite almost since its 1986 release, and now it's on Blu-ray. The film, at 25, is now older than either Sid or Nancy ever got to be. (Out on Blu-ray Dec. 27.)

Fuhu Nabi Kids Tablet


With tablets?finding their way into more and more households, there is a sensible need for a child-friendly solution. For most, giving your child free rein over an expensive device like the iPad 2?($499-$829, 4.5 stars) may not be the best idea. Not only are parents afraid of damage to their precious pads, but they should also be concerned about exposure to inappropriate content. That's where Fuhu's Nabi Kids Tablet ($199.99, sold exclusively at Toys R Us) steps in. The 7-inch? tablet features two distinct Android environments; one sheltered interface catering to kids and one nearly full featured, traditional Android interface. The Nabi is not a toy, but it is also not quite a tablet either. It packs a collection of pre-loaded apps and content, and a customizable child-safe interface, in a tablet that comes with a thick rubber bumper. But some problems with functionality and responsiveness hold the Nabi tablet back.??

Design and Display
The first thing you will notice about the Nabi is its striking red, oversized rubber bumper. This thing is really solid and looks like it could withstand most anything a child might throw at it (or throw it at). But the bumper isn't integrated into the tablet; it's just a peel-off gel case. It's too easily removable and doesn't make the tablet waterproof. It does offer solid drop protection, but it also makes the Power and Volume buttons difficult to press.

Under the rubber bumper is an unassuming white plastic slab. Together with the bumper, the Nabi measures 10.6 by 7.7 by 3.5 inches (HWD) and weighs 1.2 pounds; bigger and heavier than the Kindle Fire?($199, 4 stars) at 7.5 by 4.7 by .45 inches (HWD) and 14.6 ounces, but far better equipped for the rigors of playtime. The Nabi sports a 7-inch, 800-by-480-pixel TFT display, a low resolution you're more likely to find on a phone than a tablet. It's an upgrade over child-oriented tablets like the LeapFrog LeapPad Explorer's ($99, 4.5 stars) 480-by-272-pixel display, but not as nice as the 1,024-by-600 screens on high-quality 7-inch tablets like the the Kindle Fire or Nook Tablet?($249, 4 stars). The screen is bright, and viewing angles are pretty narrow. The biggest problem with the Nabi's display is unresponsiveness. In my tests, it often took multiple touches to get the desired response. This was while navigating through the tablet's interface, and playing games like Cut the Rope.

The tablet is equipped with a mini HDMI port, so you can connect it to an HDTV for playback, and a front-facing camera. For connectivity, there's 802.11b/g Wi-Fi and Bluetooth.

OS, Apps, Performance
The real draw of the Nabi Kids Tablet is the kids-only interface it lays on top of the now-obsolete Android 2.2. Kidz Mode is a child-safe, stripped-down environment that basically consists of oversized app icons, as well as shortcuts to Nabi's child-friendly Web and multimedia content. Any parent worried about what kind of trouble their kids can get into will appreciate the walled-off nature of Kidz Mode.

Parents can customize what apps appear in Kidz Mode, deciding for themselves what is or is not appropriate for their child. The tablet comes preloaded with games, educational apps, and childrens' books and videos. You get Angry Birds and Cut the Rope, 30 preloaded childrens' books with audio tracks for reading along, and Fooz Kids University, which is math training software for elementary schoolers. Kidz Mode also features a section of kid-safe Web shortcuts to third-party sites like Cartoon Network.

Though the browser supports Flash, which is used heavily in the third-party websites, the games and content didn't work very well on the Nabi in my tests. The sites were clearly meant to be enjoyed on a real computer, with little mobile, let alone tablet, optimization. For instance, one Cartoon Network link leads to a Flash-based game, which loads properly, but requires keyboard and mouse controls to work, which the Nabi lacks.

Toyota to recall 41,000 Etios, Liva models in India

The company, which is present in India through a joint venture with the Kirloskar Group -- Toyota Kirloskar Motor, however, insisted it is not a recall but a "safety campaign".

"It is a proactive approach by us. This development is not at all dangerous but as per Toyota's worldwide policy, we are informing customers that they can come to dealers at their convenient time and replace the pipe," TKM Deputy Managing Director - Marketing Sandeep Singh said.

The company will be replacing filler hose (inlet pipe to the fuel tank) as it could crack up leading to fuel leakage when the tank is filled to the brim.

He said a total 41,000 units of Etios and Liva produced till October 8 this year since their launches will be covered under this exercise.

"It will not take more than 30-45 minutes. This is not recall, it is a safety campaign," Singh insisted.

TKM had in December last year launched the Etios sedan in the country priced between Rs 4.96 lakh and Rs 6.86 lakh (ex-showroom Delhi).

In June this year, the company rolled out its much anticipated small car Liva here, priced between Rs 3.99 lakh and Rs 5.99 lakh (ex-showroom, Delhi).

Singh said TKM is using social media such as Facebook and the company's own website to inform customers about the development.

In 2008 Toyota had announced a recall of around 20,000 units of its popular multi-utility vehicle, Innova, in India to repair a defective part that could lead to oil leakage.

The auto major has also invested Rs 3,200 crore to set up its second manufacturing facility in Bangalore, where the cars are being produced.

The company is investing Rs 1,650 crore to ramp up its production capacity by one lakh units and for increasing localisation of components by 2014.

In October this year, the company had also revealed its plans to start exports of the two models to South Africa by March next year.

Besides, Etios and Liva, Toyota also sells premium sedan Corolla Altis and Camry in the Indian market along with SUVs Fortunner and Prado.

Girl's plight deepens Israel debate on zealot Jews (Reuters)

JERUSALEM (Reuters) ? Prime Minister Benjamin Netanyahu stepped up pledges to curb Jewish zealotry in Israel on Sunday after an 8-year-old girl complained of being menaced by ultra-Orthodox men who deemed her dress immodest.

While his conservative government insists such incidents are fringe phenomena in the mostly secular country, Netanyahu's repeated announcements on the matter reflected concern about widening religious and political schisms.

"In a Western, liberal democracy, the public realm is open and safe for all, men and women both, and neither harassment nor discrimination have any place there," he told his cabinet in broadcast remarks.

Netanyahu said he had ordered law enforcement authorities to crack down on "whoever spits, whoever lifts a hand (in violence), whoever harasses" and to remove street signs segregating men from women in some ultra-Orthodox districts.

The statement appeared to have been prompted by an expose on Israel's top-rated weekend news about intra-Jewish friction in Beit Shemesh, a town of about 87,000 people near Jerusalem.

Naama Margolese, 8, told Channel Two television she was terrified of walking to her moderate Orthodox school because of passersby who want her "to dress like a Haredi" - the Hebrew term for the ascetic, black-coated Jews who are in "awe" of God.

"I'm afraid I might get hurt or something," the girl said.

Margolese's mother Hadassa, an American immigrant who wore a headscarf and skirt in deference to religious Jewish tradition, said the sidewalk abuse could include spitting, curses like "whores" and "bastards" and calls to "clear out of here."

"If that's what happens now, and they (authorities) don't do anything, what will happen in another few years?" she told Israel's Army Radio on Sunday. "This is a terrorist group."

Returning to Beit Shemesh on Sunday, a Channel Two crew was mobbed by ultra-Orthodox Jews who stoned their car, wounded a reporter and stole equipment, police said. The crew was rescued by police, who said they were questioning suspected assailants.

WIDE MARGIN

Separately, police said they had arrested a Beit Shemesh man for spitting at a woman, and that he could face assault charges.

In the report broadcast on Friday, Channel Two showed a Beit Shemesh street sign instructing women to keep to one side, away from a synagogue. A few ultra-Orthodox men who agreed to be interviewed sought to justify their forcible occlusion of women.

Israeli media have debated the impact of religious gender segregation on public transport and the conscript army, where some pious troops prefer to shun female instructors and singers.

The ultra-Orthodox make up only about 10 percent of Israel's population of 7.7 million. But their high birthrates and bloc voting patterns have helped them secure welfare benefits and wider influence. One of Netanyahu's biggest partners in the coalition government, Shas, is a party run by rabbis.

According to Channel Two, its Beit Shemesh story has generated momentum for a demonstration against ultra-Orthodox coercion in the town, scheduled for Tuesday.

Moshe Abutbol, the Shas mayor of Beit Shemesh put the number of townspeople involved in the abuse at between 20 and 50.

Emulating Israeli commandos who disguise themselves as Arabs for missions in Palestinian areas, the police have officers who work undercover among the ultra-Orthodox. Israeli television showed them raiding a religious protest in Jerusalem on December 17.

Netanyahu has also condemned radical Jews behind a spree of vandalism against Palestinian property and Israeli garrisons in the occupied West Bank, attacks designed to bog down government moves to raze settlements built without a permit.

Facebook, Google, Yahoo Executives Summoned over Offensive Content

Senior managers from 19 Internet companies and Web sites, including Facebook, Microsoft, Google, Yahoo and YouTube, have been summoned for allegedly hosting offensive content, media reports said on Friday.

According to the terms of a complaint, filed by Vinay Rai, a journalist, a Metropolitan Magistrate in New Delhi, Sudesh Kumar, has directed the government to take the "immediate" and "appropriate" steps and file a further report by Jan. 13.

"It appears from a bare perusal of the documents that prima facie the accused in connivance with each other and other unknown persons are selling, publicly exhibiting and have put into circulation obscene, lascivious content which also appears to the prurient interests and tends to deprave and corrupt the persons who are likely to read, see or hear the same," the court said in its statement.

"It is also evident that such contents are continuously openly and freely available to everyone who is using the said network irrespective of their age and even the persons under the age of 18 years have full and uncensored access to such obscene contents," it added.

The accused companies have been charged for committing offences punishable under Section 292 (sale of obscene books), Section 293 (sale of obscene objects to young people) and Section 120-B (criminal conspiracy) of the Indian Penal Code.

The court noted that among the materials submitted by the complainant were obscene pictures and derogatory articles pertaining to Prophet Mohammad, Jesus Christ and various Hindu gods and goddesses. The Delhi court's order comes in the wake of uproar by Internet users over a perceived move by the government to censor online content.

A New York Times report earlier this month said that Telecoms and Information Technology Minister Kapil Sibal had called executives from Internet companies, including Facebook, Twitter and Google to remove content that maligned Congress party chief Sonia Gandhi.

The move unleashed a storm of criticism from Internet users, who complained of censorship in the world's largest democracy, who denied promoting such actions while adding that some images and statements risked fanning tensions in conservative India.?

A court in the northern German town of Osnabr?ck has ruled that a farmer is allowed to feed his pigs vast quantities of raw onions, despite complaints from neighbours about their eye-watering gaseous emissions.

The farmer has been feeding his 1,500 pigs several cubic metres of onions every week for the past 14 years, but city authorities ordered the farmer to stop, and threatened to fine him ?2,500 fine, after locals complained of the resulting pungent porcine farts.

The council justified its decision on the grounds that planning permission for the pigsty forbade ?strong-smelling foods, e.g. kitchen waste.?

But the court overturned the decision, saying that the city council had imposed the penalty on the assumption that the onions were to blame for the stink without providing sufficient evidence.

According to the court?s statement, the court called in an expert witness on ?emissions control,? who testified that ?onions did not count as strong-smelling foods, because they are untreated organic raw materials that have not decomposed.?

There was no indication, however, that the expert lived near the farm.

But the legal procedure has not been exhausted on this noxious issue. Onions could still be banned as pig food, if the council proves they are responsible for the distinctive farts.

The farmer has not been named, to protect his identity, but he is reportedly easy to find in a favourable wind.

Merck gets approval for wider use of HIV drug (Reuters)

(Reuters) ? U.S. health regulators have approved an expanded label for Merck & Co's treatment for HIV for use with other antiretroviral drugs in children and adolescents.

The safety and effectiveness of the drug was evaluated in a clinical trial of 96 children and adolescents of ages 2-18 years with HIV-1 infection, the U.S. Food and Drug Administration said in a statement.

The drug, Isentress, was first approved for use in adult patients in October 2007.

Holiday Greetings Bring Troops Closer to Home

WASHINGTON, Dec. 23, 2011 ? Flick on just about any TV or radio station in the coming days, and you?re likely to catch a broadcast of a local service member deployed or stationed overseas sending holiday wishes to loved ones and friends back home.

The Joint Hometown News Service produces thousands of these TV and radio messages each year through its popular holiday greetings program.

The format for the broadcasts is simple: service members who volunteer to participate look into the camera and give their name and location, followed by an extemporaneous holiday greeting.

Some of the troops? messages are quite emotional, Natasha Schleper, broadcast chief of the news service, said, especially those from young service members on their first deployments.

Producers edit the raw footage down to about 15 seconds before disseminating them to media outlets.

The greetings are a hit with media outlets anxious to air greetings from local service members deployed or stationed overseas. More than 1,500 TV stations and almost 3,000 radio stations subscribe to the distribution list, officials said. ESPN and several cable news outlets are among the subscribers.

Some radio and TV stations go the extra step to telephone service members? families in their communities before their loved one?s message airs to be sure they don?t miss it. Rick Blackburn, director of the Joint Hometown News Service, calls that outreach ?remarkable.?

To deliver those greetings in time for the holidays, Joint Hometown News Service crews started early and fanned out around the world. In addition to gathering greetings from service members and their families from Europe to Asia, they tap into American Forces Network resources to cover troops on the front lines.

This year, most of the greetings were recorded during a 30-day period and the raw footage was edited and uploaded on a server for subscriber access, Schleper reported.

One deployed soldier who previously was part of the hometown news team, Army Staff Sgt. Kimberly Cooper-Williams, volunteered to help again this year. She personally oversaw the taping of 430 greetings from service members throughout Kuwait within a four-day period.

The fruits of the crews? labors began showing up on radio and TV stations during Thanksgiving week.

On Nov. 23, for example, CNN?s Anderson Cooper aired several of the messages during his national daytime program.

Earlier this month, another holiday greeting from a Marine deployed to Afghanistan was aired between periods at a NHL hockey game on the coliseum's jumbotron video screens, to the surprise and delight of his attending family in the crowd.

The impact of these messages -- first on the camera crews, then on the families and audiences at home -- continues long after the messages air, Schleper said.

?That?s someone?s child, someone?s father or mother who?s away from home, not enjoying the holidays in their own living rooms with their families,? she said.

?The greetings are an important reminder to people at home, many of them caught up in the hustle-bustle of getting ready for the holidays, that as they go about their business, the troops are out there, continuing to defend the nation.?

Beyond the Light Switch Wins 2012 Alfred I. duPont-Columbia University Award

Beyond the Light Switch, a Detroit Public Television two-part documentary hosted by Scientific American Associate Editor David Biello, has been awarded?a Silver Baton 2012 Alfred I. duPont-Columbia University Award, it was announced today.
